A woman has died after a bus and two cars collided north-east of Melbourne at about 2:40pm today.

Victoria Police have said the woman was driving a white car and died at the scene of the crash, which took place on the Melba Highway near Glenburn.

A number of passengers in the bus have been hospitalised, including a teenage boy and a man in his 50s with serious injuries.

Two bus passengers are also in a serious condition and several others have been hospitalised following the crash on the Melba Highway near Glenburn, in the Shire of Murrindindi, at around 2:40pm.

Victoria Police said it understood the bus collided with two cars, including a white car whose driver died at the scene.

The driver and passenger of the second car, a blue Toyota, sustained minor injuries.

An Ambulance Victoria spokesperson said paramedics assessed 21 patients at the scene.

A teenage boy was flown to The Alfred in a serious condition with upper body injuries, while a man in his 50s in a serious condition was taken by ambulance to Box Hill Hospital.

A teenage girl and another teenage boy were also taken to hospital in a stable condition.

The ABC understands two bus passengers were ejected from the bus during the crash.

Authorities say 16 other patients did not require emergency treatment or transport.

Police believe 18 people, including the driver, were travelling in the bus at the time of the collision, which took place near Two Hills Road.

Specialist crews including an Air Ambulance Victoria helicopter and Advanced Life Support units assisted with the response.

Emergency services personnel remained at the scene at until about 4:30pm on Thursday afternoon.

Charter company Mee's confirmed the bus involved left Mansfield at about 1pm bound for Melbourne as part of a public transport service.

The ABC understands the bus driver was an experienced, long-term driver for the company, and was not seriously injured in the crash.

Vision from the scene suggests the bus crashed through a road barrier on the side of the road it had been travelling along.

At least two other vehicles appear to have been badly damaged. 

"This is a terrible incident — multiple people are injured and at least one person has died," he said in a post on X (formerly Twitter).

"Our thoughts are with everyone involved, their families and loved ones."

The section of the Melba Highway where the crash took place was ranked as the sixth most dangerous in the state in an RACV survey this year.

The highway has been closed to traffic between Castella and Yea following the collision.

Motorists are advised to detour through Kinglake and Flowerdale via Healesville-Kinglake, Whittlesea-Kinglake, and Whittlesea-Yea Roads.

Detectives from the Major Collision Investigation Unit are attending the scene.
